摘要
目的在RNA水平研究小鼠耐氨甲蝶呤细胞中亚甲基四氢叶酸还原酶(MTHFR)的表达情况。方法采用RT-PCR的方法,对小鼠氨甲蝶呤(MTX)抗性细胞系中MTHFR的表达进行分析。结果在NIH3T3、3T3R500、MTX100、M3T200中MTHFR的表达量基本相同,而在MTX300中MTHFR的表达量明显下降。结论在RNA水平,耐药细胞耐药浓度的改变可导致MTHFR表达改变。
Objective To study the expresssions of MTHFR in Methotrexate-resistant mouse cell lines. Methods The expression of MTHFR is detected by RT-PCR in the MTX-Resistant mouse cell lines. Results As concentration differs in NIH3T3 and MTX-resistant mouse cell lines, the expressions of MTHFR are different. In NIH3T3,3T3R500,MTX100 .MTX200, the expressions of MTHFR RNA are approximately the same, but it is markedly lowered in MTX300. Conclusion The expression of MTHFR RNA way be changed by high level of MTX.
