山羊活体采卵及卵母细胞体外受精研究

Ovum Pick-up and Fertilization in vitro of Follicular Oocytes for Goats

选择健康空怀山羊母羊,随机分组,分别用PMSG 1000 u和FSH 325～375 u超数排卵处理,研究利用外源激素进行超排对山羊活体采卵效果的影响,比较活体采集与屠宰场采集的卵母细胞体外受精效果.结果表明,经超排处理后,卵巢明显增大,同时提高了采集卵母细胞的数量和质量.用FSH超排,≥2 mm的卵泡数为23.00个±5.69个,采集卵母细胞数为12.67枚±4.06枚,极显著大于对照组（P＜0.01）.活体采集的卵母细胞的体外成熟率及受精率分别为44.44%和33.33%,均略高于从屠宰场获取卵巢采集的可用卵泡卵母细胞,但差异不显著（P＞0.05）.

The effect of donor does treated with pregnant mare serum gonadotrophin （PMSG） or follicular stimulating hormone （FSH） on oocyte recovery by ovum pickup （OPU） and in vitro embryo production was studied in this paper. A total of 8 does were selected and separated into three groups randomly.The experimental goats were synchronized by the means of intra - vaginal sponges and then were introduced multipleovulation by using 1000 u PMSG or 325-375 u FSH. The condition of ovary and the ovum picked - up were detected after multiple ovulation. The results showed that the ovary of treatment group were obvious growth and more follicles （≥2 mm） were collected than those of control groups. The average number of follicles at aspiration increased from 2. 50±2.12 for control group to 12, 67 ±4.06 for animals treated with FSH. More follicles were aspirated and the numbers of oocytes recovered per does increased with FSH treatment （P〈 0. 01）, but the differences were not significant between PMSG treatment and control group （P〉 0.05）. The maturation rate and cleavage rate of OPU oocytes were higher than those of oocytes collected from slaughtered goats, but no significant was found between the groups.
