摘要
使用适用于分离纯化还原型谷胱甘肽的005×7强酸型阳离子交换树脂,该树脂是一种适合分离纯化小肽的强酸性苯乙烯系阳离子交换树脂,通过L9(33)正交实验对渗透法抽提GSH与对羟基苯甲酸丙酯法进行了对比研究。渗透法抽提的最佳条件为:pH3.2,50℃保温4h,其GSH含量为63.49mg/100mL,而对羟基苯甲酸丙酯法的GSH含量仅为46.32mg/100mL。同时将两种抽提液分别用005×7离子交换柱分离纯化,浓缩冷冻干燥,产品经DTNB法测含量,其结果表明,渗透法抽提液经分离后得到的产品中的GSH含量为0.27mg/g,明显高于对羟基苯甲酸丙酯法的0.19mg/g,而且高效液相色谱图显示的峰面积渗透法也大于对羟基苯甲酸丙酯法。
This article uses the 005×7 strpmg acod catopm exchange resin for the first time to purity the Reduced Glutathione (GSH). The comparisons were made in the paper between extraotions of GSH by osmosiss and hordenine propyl formate method by orthogonal experiments. The optimum condition for osmosis extraction is :pH3.2 ,50℃ for 4h. The extraction yield is 63.49mg/100mL, but only 46.32mg/100mL was obtained by hordenine propyl formate method. These two extracts were further separated by 005×7 ionic exchange column, concentrated and freeze-dried. The content of GSH was analyzed by DTNB method. The results indicated that the final yield obtained by the osmosis process is 0.27mg/g. obviously higher than 0.19mg/g by hordenine propyl formate method. HPLC results also prove the higher extraction yield by osmosis processes.
