金雀异黄素对血小板源生长因子刺激下大鼠系膜细胞周期调节蛋白的影响

摘要 金雀异黄素(genistein)可影响多种细胞尤其肿瘤细胞的细胞周期调节蛋白的表达而调控细胞的增殖与凋亡,并通过调节细胞外基质(ECM)降解系统酶的活性影响基质的积聚。我们的前期实验已证实血小板源生长因子(PDGF—BB)可诱导体外培养的大鼠系膜细胞(MC)增殖,金雀异黄素可呈浓度依赖性抑制其增殖。既然细胞增殖的调控是在细胞周期水平上进行,而细胞周期又由细胞周期调节蛋白来调控,本实验我们拟研究金雀异黄素如何影响细胞周期调节蛋白的表达最终达到抑制细胞增殖的效应,为金雀异黄素在肾病中的应用提供实验依据。
