摘要
以人表皮生长因子为研究对象,分3段合成了hegf基因片段,运用套叠PCR技术进行连接,形成全长hegf(159bp)并克隆到酵母表达载体pGAPZα-A中.利用同裂酶技术构建串联体表达载体,获得了分别含2、3、4拷贝的串联体表达载体pGAPZα-2hegf、pGAPZα-3hegf和pGAPZα-4hegf,为下一步进行基因表达及其生物学活性分析奠定了基础.
The human epidermal growth factor(hEGF) using overlap extension PCR, then cloned into Pichia to constructed multi-copy expression vectors and 2, pGAPZα-3 hegf and pGAPZα-4 hegf were successfully for gene expression and bio-activity analysis. gene was synthesized from three fragments, connected expression vector pGAPZα-A. Isoschizomers were used 3, 4 multi-copy expression vectors pGAPZα-2hegf, constructed, respectively. The results were a foundation for gene expression and bio-activity analysis.
