缺氧诱导因子-1α和血管内皮生长因子在大肠腺瘤与非肿瘤性息肉中的表达比较

Comparison of the Expressions of Hypoxia-inducible Factor-1α and Vasculoendothelial Growth Factor in Colorectal Adenoma and Non-neoplastic Polyp

摘要 通过分析大肠腺瘤与非肿瘤性息肉中缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)的表达差异,探讨与肿瘤生成的关系。采用免疫组化SP法检测36例大肠腺瘤、36例非肿瘤性息肉、13例腺癌和11例正常黏膜组织中HIF-1α和VEGF的表达,并用CD34单抗标记各组新生血管内皮,计数微血管密度(MVD)。结果显示,大肠腺瘤组HIF-1α、VEGF的表达和MVD值均高于非肿瘤性息肉组(P〈0.05);腺瘤组HIF-1α与VEGF的表达呈显著正相关(γs=0.644,P〈0.01),MVD值与VEGF的表达强度呈显著正相关(γs=0.640,P〈0.01)。结果表明,HIF-1α通过上调VEGF的表达促进血管生成,HIF-1α和VEGF在腺瘤的高表达和非肿瘤性息肉的低表达可能是二者恶变趋势差异的重要分子调控机理之一。 The objective of the study was to analyze the difference of the expressions of hypoxia-inducible factor-1α(HIF-1α) and vasculoendothelial growth factor(VEGF) and their relationships with oncogenesis. Immunohistochemical SP method was applied in the measurement of the HIF-1α expression and VEGF expression of 36 cases with colorectal adenoma,36 cases with non-neoplastic polyps, 13 cases with adenocarcinoma and 11 cases with normal mucous tissues. The newly grown vasculoendothelium was labelled with CD34 monoclonal antibody,and microvascular density(MVD) was then counted. As results, the expressions of HIF-1α & VEGF and the value of MVD were higher in the colorectal adenoma group than in the non-ne- oplastic polyp group( P〈0.05), the expressions of HIF-1α and VEGF in the adenoma group had markedly positive correlation(γs=0. 644, P 〈0.01),the value of MVD with the intensity of VEGF expression had also markedly positive correlation(γs=0. 640, P 〈0.01). It is suggested that HIF-1α may be involved in the promotion of angiogenesis by up-regulation of the expression of VEGF, and the higher expressions of HIF-1α & VEGF in adenoma and the lower expressions of HIF-α& VEGF in non-neoplastic polyps may be one of the important molecular regulation mechanisms in malignant development tendency.
