摘要
以三色羽扇豆的叶柄为外植体,在附加不同浓度的BA、IBA、NAA和2,4-D的MS培养基上进行离体培养。结果表明:诱导三色羽扇豆叶柄形成愈伤组织的理想培养基为MS+BA 0.4 mg/L+2,4-D 1.2 mg/L;诱导愈伤组织分化和分化不定芽增殖培养的理想培养基为MS+BA 0.6 mg/L+NAA 0.2 mg/L;分化不定苗生根的理想培养基是1/3MS+IAA0.5 mg/L;试管苗移栽的理想基质是炉灰渣;露地栽培的试管苗具有抗逆性强、生长旺盛、根系发达、花期延长,冬前枯萎时间晚等特点。
The petiole of Naturtium officinale R was as explant and cultured on MS culture media contained different concentration of BA, IBA, NAA and 2,4-D. The results indicated that the optimum culture media which induced the callus was MS contained BA 0.4 mg/L and 2,4-D 1.2 mg/L; the optimum culture media which induced the callus differentiation and adventitious bud proliferation was MS contained BA 0.6 mg/L and NAA 0.2 mg/L; the optimum culture media which induced the adventitious root was 1/3MS contained NAA 0.6 mg/L and the stove ashes was substrate which was optimum for transferring test-tube plantlet. The seedlings cultured in the field had higher resistance, faster growth, more developed root system, longer flower period and wilting delay pre-winter.
