黄芪对慢性饮酒大鼠急性肺损伤的干预作用

摘要 目的研究慢性饮酒对大鼠注射油酸-脂多糖致肺损伤易感性的影响及中药黄芪的干预效果。方法SD大鼠48只,经慢性饮酒(或饮水)6周后,注射油酸-脂多糖致肺损伤,部分大鼠致肺损伤同时注射黄芪注射液,检测肝肺组织中还原型谷胱苷肽(GSH)、丙二醛(MDA)水平,以及支气管肺泡灌洗液(BALF)中蛋白含量及PaO2的变化。结果肺损伤组PaO2、肝肺组织中GSH水平下降,MDA及BALF中蛋白水平显著升高,尤其以饮酒大鼠(饮酒损伤)改变显著。与肺损伤组比较,药物干预组(饮酒损伤药物组及饮水损伤药物组)PaO2、肝肺组织中GSH水平升高,MDA及BALF中蛋白水平下降,尤以饮酒损伤药物组改变显著。结论慢性饮酒史可加重创伤后严重感染大鼠的氧化、抗氧化失衡,加重急性肺损伤的程度,黄芪注射液可改善肺损伤尤其是慢性饮酒大鼠肺损伤时的氧化抗氧化失衡,然而不良反应不容忽视。 Objective To investigate the influence of chronic alcohol ingestion on the susceptibility of acute lung injury induced by injection of oleic acid ( OA) and hpopolysaccharide( LPS), and the intervention effect of Astragalus membranaceus milkvetch root. Methods After having ingested alcohol for 6 weeks, SD rats were injured with OA and LPS to develop the acute lung injury. Some of the injuried rats were given Astragalus injection. PaO2 and protein content in the bronchoalveolar lavage fluid(BALF), and GSH, MDA levels in the lung and liver tissues were observed.Results PaO2 and GSH decreased, MDA and protein content in BALF increased in the lung injury group, especially in alcohol group. While in the drug intervention group,the above- mentioned indexes ameliorated,especially in alcohol group. Conclusions Chronic alcohol ingestion aggravates the imbalance of oxidation/antioxidation and severity of the lung injury in the rat. Astragalus improves the imbalance especially in the chronic alcohol ingestion rat.
