摘要
利用土豆组织中丰富的多酚氧化酶,将土豆切片与氧电极配合,制成了植物组织膜儿茶酚(邻苯二酚)电极。在2.2~88μg/ml邻苯二酚浓度范围,电极的动力学响应与邻苯二酚浓度间呈线性关系,相关系数0.9998。研究了介质条件、温度、切片厚度、固定化等条件的影响,测定了电极的重现性、选择性和土豆切片的活性期限,计算了该条件下酶反应的Michaelis常数。采用动力学方法测得10份样品的平均回收率为100.2%,标准偏差为1.07%。
A tissue section electrode sensitive to catechol has been made with a thin slice of potato coupled with an oxygen electrode. The dynamic response of electrode is linearly related with the concentration of catechol over the range of 2. 2~88μg/ml and the correlation coefficient is 0. 9998. Important experimental variables including the effect of buffer composition, operation pH, temperature, tissue slice thickness and immobilization of enzyme in the tissue were investigated. The reproducibility, selectivity and lifetime of tissue membrane electrode were determinated, The Michaelis constant of enzyme reaction at present condition was calculated. The average recovery for ten samples determined by the dynamic response method was 100. 2% with a standard deviation of 1. 07%.
