碱性磷酸酶及其免疫标记物活性的荧光法测定 被引量:1

FLUOROMETRIC MEASUREMENIT OF ALKALINE PHOSPHATASE ACTIVITY AND ITS APPLICATION TO IMMUNOASSAY

摘要 为提高碱性磷酸酶(ALP)活性测定的灵敏度,对以4-甲基伞形酮磷酸酯(4-MUP)为底物的荧光法进行了研究。结果ALP的线性范围达1.7×10-5~1.7×10-8IU/ml,检出限为4.0×10-8IU/ml;用本法测定了20份人血清样品,误差为3.5%;并测定了免疫标记物人IgG-ALP活性,发现其活性与荧光强度也呈线性关系。研究表明,通过纯化4-MUP,优化分析条件,其灵敏度和实用性都得到明显提高。 In order to improve the sensitivity of detection of alkaline phosphatase(ALP)activity,a fluorometric method based on 4-methylumbelliferyl phosphate(4-MUP)as substrate was investigated.The linear range of ALP was 1.7×10 -5 ~1.7×10 -8 IU/ml,and the detection limit for ALP was 4.0×10 -9 IU/ml.Twenty samples of human serum were used(the relative error was 3.5%).Human IgG-ALP was also investigated and its concentration was linear with fluorescence intensity too.The results showed that the sensitivity was greatly improved by puring 4-MUP and optimizing the test conditions.
