摘要
目的检测胆管癌和癌旁胆管组织中Survivin蛋白和mRNA的表达,探讨其在胆管癌发生发展中的作用。方法应用免疫组化SP法检测48例胆管癌和其中30例癌旁胆管组织中Survivin蛋白的表达,同时应用RT-PCR技术检测这些组织标本中Survivin mRNA的表达,并与临床病理资料进行相关性分析。结果48例胆管癌组织中,Survivin蛋白阳性表达率为79.2%(38/48),在癌旁胆管组织中Survivin蛋白阳性表达率6.7%(2/30),两者比较差异有显著性意义(P<0.05)。48例胆管癌组织中,有30例Survivin mRNA的表达高于配对正常组织,胆管癌组织中Survivin mR-NA的表达水平(2.18±1.21)明显高于正常对照组织(1.01±1.03)(P<0.05)。临床病理资料的相关性分析显示,胆管癌组织中Survivin蛋白和mRNA的表达分布与临床病理特征无相关性。结论Survivin基因转录和蛋白表达可能参与胆管癌的发生发展过程,检测Survivin表达可能有助于阐明胆管癌的发生机制。
Objective To study the expression of Survivin protein and gene in extra-hepatic cholangiocarcinoma and to elucidate its role. Methods The expression of Survivin protein and gene was detected by SP immunohistochemical technique and RT-PCR in 48 cases of cholangiocarcinoma, 30 cases of adjacent histologically noncancerous bile duct respectively. The correlation between the expression of Survivin protein and mRNA and clinicopathological parameters in extra-hepatic cholangioncarcinoma was analyzed. Results Survivin was expressed in 38 of 48 cholangiocarcinomas (79.2 %). In contrast, the expression rate of Survivin in adjacent histologically noncancerous bile ducts was 6.7% (P〈0.05). The expression level of Survivin mRNA in tumor tissues (2.18±1.21) was significantly higher than that in normal tissues (1.01±1.03, P〈0. 05). No correlation was found between the expression of Survivin protein and gene and clinical features. Conclusion Survivin gene transcription and protein expression are mostly likely involved in the proliferation and malignant transformation of bile epithelia and then malignant progression of bile duct carcinomas. The detection of Survivin expression may elucidate the carcinogenesis of bile duct.
