摘要
目的研究大鼠糖尿病性白内障(diabetic cataract,DC)晶状体上皮细胞(lens epithelial cells,LECs)水通道蛋白-1(aquaporin-1,AQP1)表达水平的变化。方法链脲佐菌素(streptozotocin,STZ)60 mg/kg体重一次性尾静脉注射制作大鼠糖尿病模型,分别在造模后不同时间点摘取模型组及对照组大鼠的眼球,制作石蜡切片,应用免疫组织化学方法和计算机图像分析技术对晶状体囊膜下上皮细胞AQP1进行检测。结果透明晶状体和DC各期晶状体囊膜下上皮细胞膜均有AQP1阳性表达。图像分析显示:在造模2周糖尿病大鼠晶状体尚透明,LECs的AQP1平均吸光度(A)值即下降,与对照组比较差异有显著性意义(P<0.05);DC各期LECs的AQP1表达均显著低于对照组(均P<0.01),随着白内障的发展,A值逐渐下降。除DCⅡ期与Ⅲ期之间A值差异无显著性意义外,其余DC各期之间差异均有极显著性意义(均P<0.01)。结论LECs胞膜AQP1表达量减少先于DC的发生,DC早期AQP1的表达量显著下降,AQP1表达改变可能在DC发生发展中起一定作用。
Objective To observe the expression change of aquaporin 1 (AQP1) in lens epithelial cells (LECs) of rat diabetic cataract (DC). Methods An animal model of diabetes was prepared by intravenous injection of streptozotocin (60 mg/kg body weight) in 2-month-old rats. The eyeballs were extracted periodically from each experimental group and control group and made into paraffin-embedded sections. Immunohistochemical techniques and computer-imaging analysis were used to localize and semi-quantify the expression of AQP1 in LECs. Results The expression of AQP1 was positive on the membrane of normal and diabetic LECs. The expression of AQP1 was decreased significantly in two-week group. The expression of AQPI in LECs of DC rats was lower than in clear lens (P〈0.01). The expression of AQP1 was gradually decreased with DC development. There was statistically significant difference between each stage of DC (P〈0.01) except DC Ⅱ and DC Ⅲ (P〉0.05). Conclusion The expression of AQP1 in LECs was decreased prior to the occurrence of DC. The expression of AQP1 was decreased significantly in the early stage of DC. The expression change of AQP1 may play a partial role in the occurrence and development of DC.
