摘要
目的:观察β-细辛醚对PC12细胞和乳鼠皮层神经细胞形态学及细胞活力的影响。方法:用免疫细胞化学方法检测皮层神经细胞特异性标志物NSE、GFAP的表达;在体外将不同浓度β-细辛醚与PC12细胞和皮层神经细胞各作用24 h,用相差显微镜观察细胞形态的改变,并用MTT比色法检测细胞活力的变化。结果:原代培养的乳鼠皮层神经细胞大多数NSE呈阳性表达,少量GFAP呈阳性表达;β-细辛醚与PC12细胞作用24 h,低浓度β-细辛醚(7.5、15、30、60μg/ml)有促增殖作用,高浓度β-细辛醚(120、140、2480μg/ml)则可抑制其增殖,且随药物浓度的增加而增强;β-细辛醚与皮层神经细胞作用24 h,7.5、15、30、60、120μg/mlβ-细辛醚对其细胞形态和活力无明显影响,240μg/mlβ-细辛醚对其有促增殖作用,而480μg/mlβ-细辛醚对其则有损伤作用。结论:β-细辛醚可能有抗肿瘤和保护神经细胞的作用。
Objective: To observe the effects of β-asarone on the morphology and cell viability in PC12 cells and cultured neonate rat cortical neurons. Methods: The cultured neonate rat cortical neurocytes were stained immunocytochemically with NSE, GFAP antibodies, respectively; Morphological changes were observed under phase contrast microscope after PC12 cells and cultured rat cortical neurons with β-asarone of different concentrationfor 24h in vitro, and the cell viability of PC12 and cortical neurons were examined by MTT assay. Results: Most of the cultured neonate rat cortical neurocytes were positively stained with NSE antibody, and positively with GFAP in a less degree; Treatment of PC12 cells with concentrations of 7. 5,15,30,60 μg/ml β-asarone for 24h could facilitate the proliferation in PC12 cells, 120,240,480μg/ml β-asarone could inhibit the proliferation inversely, and with the concentration of β-asarone increasing, the inhibition was enhanced; Treatment of cultured neonate rat cortical neurons with concentrations of 7.5,15,30, 60,120 μg/ml β-asarone for 24h, there were no visible effects on morphology and cell viability, 240 μg/ml β-asarone could facilitate the proliferation obviously, but 480 μg/ml β-asarone induced injury on neurons. Conclusion : β-asarone maybe has anti-tumor and protective effects on cultured neurons.
