摘要
建立了尿中多胺的气相色谱测定法。取晨尿5ml,经酸水解,硅胶柱预处理、TFAA衍生化反应在,SE-52毛细管开管柱上完成尿中多胺的气相色谱测定。尿肌酐为内标,以多胺/肌酐比值为评价标准。方法回收率:腐胺96.7%,精眯102.6%,精胺98.7%。最小检出量:腐胺2ng/μl,精眯5ng/μl,精胺25ng/μl。5次重复分析结果的算术平均值是1.949(精脒μg/mg肌酐),平均值的标准偏差是S_x^-=0.041。正常人尿中多胺总量为2.01,癌症患者尿中多胺总量为44.74μg/mg肌酐。二者存在显著性差异。
The quantitative analysis of polyamines〔putrescine (Put), spermidine(Spd) and sperimine(Spm)〕in urine of the normal human and cancer patient by capillayg as chromatography is compared. Only 5ml of morning urine is acided and hydrolyzed for 5h at 100℃. After acid hydrolysis, sample is cleaned up by adsorption onto silicon gel and converted into their TFAA derivative. Creantinine in human urine is taken as internal standard. The recovery rate of polyamines is 96.7%(Put), 102.6%(Spd) and 98.7%(Spm). The standard devation(SD)of the method for Spd is 1.949+0.041(μg/mg creatinine, mean+SD). The results show that the mean level of polyamines in cancer patients urine is higher than that in the normal urine. The mean of total polyamine values in the normal urine is 2.01 and 44.74 in the cancer patients (μg/mg creatinine).
