摘要
本文介绍了用反相高效液相色谱分析油菜籽蛋白质水解产物中氨基酸组成的方法。氨基酸通过柱前衍生化反应转变成丹磺酰氯(D_ns-Cl)的衍生物,洗脱后用紫外检测器(波长254nm)进行检测。17种氨基酸及副反应产物在45min内得到良好的分离。方法的检出极限为50pmol。
A rapid and simple HPLC method for the determination of amino acids in hydrolysate of rapeseed protein is presented. The amino acids are derivatized with dansyl chloride (Dns-C1), separated on Lichrosorb RP-18 column and detected at 254nm. Complete separation of 17 amino acids was achieved in 45 min. The detection limits are less than 50pmo1.
