NT-3基因转染大鼠骨髓间充质干细胞的实验研究 被引量:4

摘要 目的检测神经营养素-3(NT-3)基因转染骨髓间充质干细胞(BMSCs)后的表达情况。方法采用密度梯度离心法获取SD大鼠BMSCs,体外培养、传代。取第4代细胞,用脂质体介导法将NT-3真核重组表达质粒plEGFP-NT-3转染BMSCs,G418筛选获得阳性细胞,用免疫细胞化学法检测NT-3转染效果。结果在阳性细胞中能够检测到NT-3基因表达,但细胞形态并没有因为NT-3的转入而有明显改变。结论通过质脂体介导法可将NT-3基因转入BMSCs中,并能稳定表达。
