pEBFP/FP6共表达质粒转染神经干细胞的实验研究

摘要 目的:将α2巨球蛋白cDNA片段(FP6)-增强型绿色荧光蛋白(pEBFP)共表达质粒pEBFP/FP6转染至神经干细胞(NSC)中,观察其对NSC生长、分化、增殖的影响。方法:培养NSC,采用脂质体转染方法将pEBFP/FP6共表达质粒转染进NSC中,利用其携带的绿色荧光和RT-PCR观测其转染情况。结果:经鉴定所培养的细胞是NSC,采用脂质体转染方法成功的将pEBFP/FP6双表达质粒转染进NSC。结论:脂质体转染能将pEBFP/FP6共表达质粒转染进NSC中,pEBFP/FP6基因对神经干细胞生长、分化、增殖无明确影响。 Objective fluorescent protein ( pEB :To study the transfection of alpha -2 macroglobulin cDNA (FP6) - enhance green FP) coexpression plasmid (pEBFP/FP6) into neural stem cell (NSC) , and observe whether FP6 - pEBFP could influence the growth, proliferation and differentiation of NSC. Methods: NSCs of rats were isolated and cultured. The pEBFP/FP6 coexpression plasmid was transfected into NSC by liposome and affirmed by utilizing green fluorescent protein and RT - PCR. Results: Cultured NSCs were identified by immunohistochemistry. The pEBFP/FP6 plasmid was transfected into NSC by liposome. Conclusion: The pEBFP/ FP6 coexpression plasmid can be transfected into NSC by liposome. After transfection of pEBFP/FP6, the growth, proliferation and differentiation of NSC were uninfluenced.
