摘要
目的:建立高效液相色谱-荧光法检测丙泊酚血药浓度的方法。方法:采用饱和硫酸铵溶液作蛋白沉淀剂,以乙腈-水(70:30,含0.1%三氟乙酸)为流动相,流速1.0ml·min-1,荧光检测器检测。结果:丙泊酚血药浓度范围为0.1~10.0mg·L-1,(r=0.999 5);最低检测限为0.02mg·L-1;平均提取回收率为87.03%,RSD为2.54%;平均方法回收率为105.08%,RSD为3.81%;日内差异RSD为4.31%;日间差异RSD为7.54%。结论:本检测方法操作简便、灵敏度高、专一性强、结果可靠,可用于丙泊酚血药浓度监测和药物代谢动力学研究。
Objective: To develop a HPLC method for determination of propofol in human plasma. Methods: Saturated ammonium sulfate was used as deproteinsation solution to prepare sample. 20μl of supernatant was injected. The mobile phase was composed of acetonitrile-water (70:30, containing 0.1% trifluoroacetic acid) as rate of 1.0 ml. min^-1. The fluorescence detector was used. Results: Propofol had a good linear relationship within 0.1-10.0 mg· L^-1 (r = 0.999 5). The limit of detection was 0.02 mg·L^-1 with an injection volume of 20μl. The extracting recovery of propofol was 87.03%, and the method recovery was 105.08%. The RSD of within-day and between-day were 4.31% and 7.54% respectively. Conclusion: The proposed method is simple, sensitive, accurate, reliable, and can be used for blood concentration monitoring and pharmacokinetic studies of propofol.
