A20的泛素修饰抑制NF-kB活性 被引量:3

摘要 转录因子NF—κB的失控能介导肿瘤和多种炎症性疾病,包括锌指蛋白A20在内的几种蛋白质紧密的控制着它的活化。近来A20针对前炎症反应细胞因子肿瘤坏死因子(TNF)、多种Toll样受体(TLR)介导的NF—κB的活化的下调机制的研究取得进展。在TNF—α途径:A20发挥两个相对的活性,相继的TNF受体相互作用蛋白(RIP)的去泛素化和泛素化,从而靶向蛋白酶体导致RIP降解,抑制了抑制性κB激酶(IKK)的活化,从而终止了TNF诱导的NF—κB的活化;在TLR途径:A20通过其N末端卵巢肿瘤(OTU)域从肿瘤坏死因子受体相关因子6(TRAF6)去除泛素链,从而终止了TLR诱导的NF—κB的活化。
