结肠癌组织中趋化因子受体CXCR4和基质金属蛋白酶MMP-2的表达及意义 被引量:5

摘要 目的观察趋化因子受体CXCR4和基质金属蛋白酶MMP-2的表达与结肠癌各项临床指标间的关系。方法采用免疫组织化学S-P法检测64例Ⅰ-Ⅳ期结肠癌组织石蜡包埋标本和相应癌旁正常结肠组织标本中CXCR4、MMP-2的表达。结果64例癌组织中37例呈CXCR4阳性,39例呈MMP-2阳性;与癌旁正常组织比较均有统计学意义;CXCR4、MMP-2表达均只与淋巴结转移与否有关,而与浸润深度、分化程度、临床分期等无关;CXCR4的表达与MMP-2的表达呈正相关。结论CXCR4、MMP-2可能与结肠癌的淋巴结转移有关,且二者在结肠癌淋巴结转移过程中可能存在某种调控或协同作用,联合检测CXCR4、MMP-2可预测结肠癌淋巴结及远处转移情况并指导临床用药。 Objective To investigate the relationship between the expressions of chemokine receptor CXCR4 and matrix metalloproteinase MMP-2 and the clinic pathological diameters in human colon cancer. Methods Immunohistochemistry was used to detect the expressions of the CXCR4 and MMP-2 in 64 cases with colon cancer. The relationship between the CXCR4 and MMP-2 expressions and the clinic pathologi- cal diameters was statistically analyzed. Results The positive rate of CXCR4 expression with lymph node metastasis was higher than that without lymph node metastasis(68.3% vs 39.1% ,P 〈 0 05 ). The positive rate of MMP-2 expression with lymph node metastasis was also higher than that without lymph node metastasis (87.8% vs 13.0% , P 〈 0 05 ). The positive expression of CXCR4 was consistent with the positive expression of MMP-2, and the expression levels of CXCR4 and MMP-2 were only correlated with lymph node metastasis, but not with TNM stages, cellular differentiation and invasion depth. Furthermore, there exists a positive correlation between the expression of CXCR4 and the expression of MMP-2. Conclusions CXCR4 and MMP-2 expressions may have some common working mechanism which mav serve as parameters for the lymphatic metastasis of colon cancer.
