摘要
目的研究大孔吸附树脂提取麦考酚酸(mycophenolic acid)的工艺条件。方法比较了D290,D303,D312,HZ802 4种大孔吸附树脂对发酵液中的麦考酚酸吸附性能,并研究了动力学过程,同时对发酵液中的麦考酚酸在D312吸附柱上的动态吸附解吸过程进行了研究。结果同其他树脂相比,大孔吸附树脂D312能更好地从发酵液中提取麦考酚酸,其吸附量在28400 mg.L-1左右,用体积分数85%乙醇和稀盐酸可以将吸附在树脂柱上的麦考酚酸有效解吸,解吸收率达95%。结论大孔吸附树脂D312是从发酵液中分离和提纯麦考酚酸的一种适宜吸附剂,该工艺简捷,并可向工业化发展。
OBJECTIVE To separate mycophenolic acid by macroporous resin. METHODS The adsorption for mycophenolic acid by four macroporous resins of D303, D290,HZ802 and D312 was compared, and adsorption dynamics process of resins was studied. And the dynamic adsorption and desorption process of mycophenolic acid within fermentation broth on D312 adsorption resin was studied. RESULTS D312 resin provide the optimum performance for the purification of mycophenolic acid among 4 macroporous resins. The adsorptive capacity of D312 resin for mycophenolic acid within fermentation broth was 28 400 mg·L^-1 , repectively, Mycophenolic acid adsorbed on the D312 resin was desorbed by 85% ethanol and watery hydrochloric acid. The average yield of desorbing mycophenolic acid reached 95%. CONCLUSION D312 resin is an ideal macroporous adsorbent for separating and purificating mycophenolic acid from fermentation broth. The investigation is satisfied and provides a new thought for the industrial production of mycophenolic acid.
