摘要 目的:通过应用HPLC—ECD分离测定猕猴血清中单胺类递质,验证其与经前期综合征(PMS)肝气逆证的内在关系,筛选科学有效的检测方法。方法:采用ZORBA×SB-C18色谱柱(4.6imm×250mm,5μm),流动相:乙酸钠100mmol·L^-1,柠檬酸85mmol·L^-1,正二丁胺0.4mmol·L^-1,辛烷基磺酸钠1.5mmol·L^-1,乙二胺四乙酸(EDTA)0.2mmol·L^-1,甲醇18%,抽滤。流速0.9mL·min^-1,量程1μA。结果:猕猴血清中去甲肾上腺素(NE),肾上腺素(E),多巴胺(DA)和5-羟色胺(5-HT)回收率依次为97.0%,97.8%,99.5%,100.3%,RSD0.22%-0.93%,重复性较好。结论:该方法操作简便、快速、准确,是检测猕猴血清中单胺类递质检测的一种理想方法。 Objective: To develop an HPLC-ECD for the determination of monoamine transmitters in serum of macaque. Method: The analysis was carried out on a ZORBAX SB-C18 column (4. 6 mm× 250 mm,5 μm) eluted with a mobile phase of metha- nolwater ( 18: 82) at a flow rate of 0. 9 mL · min^-1. Result: The recoveries of NE, E, DA, 5-HT were 97.0%, 97. 8%, 99. 5%, 100. 3%, RSD was 0. 22%-0. 93%, and the repeatability was good. Conclusion: The results prove that the method is simple, fast, accurate and can be used to determine simultaneously the concentration of monoamine transmitters in serum of macaque.
