摘要
目的:建立膝沟藻毒素(gonyautoxin,GTX)2和3的毛细管电泳分析方法。用毛细管电泳法分析和定量GTX免疫原。方法:用磷酸氢二钠-柠檬酸为运行缓冲液的区带毛细管电泳紫外检测法,对GTX2和3进行检测。结果:GTX2在0.5—118.0μmol/L,GTX3在0.5—39.0μmol/L时,有良好的线性关系。GTX2最低检测限为0.38μmol/L,GTX3最低检测限为0.36μmol/L(S/N≥3)。日内精密度为:GTX2是2.5%,GTX3是2.6%;日间精密度是:GTX2为5.6%,GTX3为6.3%。用该法定量GTX2&3免疫原中GTX2的浓度为325.8±32.8μmol/L,GTX3的浓度为287.6±15.1μmol/L。结论:用磷酸氢二钠-柠檬酸为运行缓冲液的高效毛细管电泳法,可对GTX2&3相关样品进行快速而又灵敏的分析和定量。
Objectives:To establish a method to determine gonyautoxin (GTX) 2&3 by capillary electrophoresis, Methods: This assay was based on the ultraviolet absorbance of GTX 2&3 at zone capillary electrophoresis. The running buffer was disodium hydrogen phosphate - citric acid. Results:The linearity of GTX2 was from 0, 5 μmol/L to 118.0 μmol/L, and GTX3 from 0. 5 μmol/L to 39. 0 μmol/L. The minimal detection limit of GTX2 was 0. 38 μmol/L, and GTX3 was 0. 36 μmol/L(S/N≥3). With- in - run precision of GTX2 was 2. 5%, and GTX3 2. 6%, Between - run precision of GTX2 was 5. 6% ,and GTX3 6. 3%. Concentration of immunogen GTX2 was quantified as 325.8 ± 32, 8 μmol/L, and GTX3 quantified as 287.6 ± 15.1 μmol/L. Conclusion:GTX 2&3 can be quantified rapidly and sensitively by high performance capillary electrophoresis in disodium hydrogen phosphate - citric acid buffer system.
