摘要
目的进行桔梗药材的质量标准研究。方法以桔梗皂苷D为对照品,采用TLC法进行定性鉴别,HPLC法进行含量测定,Zorbax C18柱(4.6mm×150mm,5um),流动相为甲醇-乙腈-0.2%磷酸水溶液(3:25:72),检测波长为204nm,柱温为40℃,流速为1.0mL/min。结果桔梗皂苷D在0.226-2.486ug线性关系良好(r=1.0000),平均回收率101.81%,RSD1.70%。结论通过10批次不同产地桔梗的鉴别和含量测定,证明本方法灵敏,准确可靠,重现性好,可用于桔梗药材的质量控制。
Objective To establish a quality standard of Platycodon grandiflorus. Methods The presence of platycodin D were identified and assayed by TLC and HPLC, respectively. Extracts were separated on a Zorbax C18 column with methanol-acetonitrile-0.2% phosphoric acid (3: 25: 72) as mobile phase. The detection wavelength was set at 204 nm. The column temperature was 40 ℃ and flow-rate was 1.0 mL/min. Results The linearity was obtained in the range of 0. 226 -2. 486 ug( r = 1. 000 0). The average recovery rate was 101.81% ( RSD = 1.70% ). Conclusion The methods were sensitive, accurate, reliable, and suitable for the quality control of Platycodon grandiflorus.
