摘要
目的:建立同时测定藏药湿生扁蕾中木犀草素和1,7-二羟基-3,8-二甲氧基(口山)酮的含量测定方法。方法:采用Hypersil BDS-C_(18)色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇(A)-0.4%磷酸水溶液(B)梯度洗脱,流速为1.0 mL·min^(-1),检测波长为260 nm。结果:木犀草素在1.2~48μg·mL^(-1)范围内线性良好,回归方程为 Y=54.311X-31.725(r=0.9993),平均回收率为96.3%;1,7-二羟基-3,8-二甲氧基(口山)酮在2.4~96μg·mL^(-1)范围内线性良好,回归方程为 Y=109.9X-111.99(r=0.9999),平均回收率为97.5%。结论:该法简便,准确,重现性好,适用于湿生扁蕾中木犀草素和1,7-二羟基-3,8-二甲氧基(口山)酮的定量分析。
Objective:To establish an HPLC method for the determination of luteolin and 1,7 - dihydroxy - 3, 8- dimethoxyxanthone in Gentianopsis paludosa Ma. Methods: A Hypersil BDS -C18 column (4.6 mm × 250 mm,5 μm)was used with gradient elution the mobile phase of methanol -0.4% phosphoric acid and detected at 260 nm. The flow rate was 1.0 mL · min^-1. Results:The calibration curves was linear in the range of 1.2 -48 μ g · mL^-1 for luteolin,the regression equation being Y = 54. 311X - 31. 725 ( r = 0. 9993 ) , and the average recovery was 96.3% ;The calibration curves was linear in the range of 2.4 -96μg · mL^-1 for 1,7 - dihydroxy - 3,8 - dimethoxy xanthone, the regression equation being Y = 109.9X - 111.99 ( r = 0. 9999 ) , and the average recovery was 97.5 %. Conclusion: The method is simple, accurate and reproducible for quality control of Gentianopsis paludosa.
