外周血造血干细胞动员后采集时机的快速动态监测 被引量:1

摘要 目的探讨一种简单、快速判断外周血造血干/祖细胞最佳采集时机的方法。方法利用Sysmex XT2000I多通道血细胞分析仪的VCS和化学染色技术,对9例异基因外周血造血干细胞(Allo-PBSCT)的健康供者和5例自体外周血造血干细胞(auto-PBSCT)患者,在动员过程中采集外周血进行外周血循环幼稚细胞(PBCIC)检测。同时应用流式细胞术(FCM)和集落培养法分别对上述外周血中的CD34+细胞、粒-巨噬细胞集落形成单位(CFU-GM)进行检测,观察三者之间的相关性。结果PBCIC与CD34+细胞及CFU-GM呈良好的正相关,相关系数分别为0.789和0.877。健康供者干细胞动员后PBCIC的峰值出现于使用G-CSF后的第5-6天,PBCIC的变化幅度较CD34+和CFU-GM的变化幅度大。结论利用Sysmex XT2000I多通道血细胞分析仪检测PBCIC数能快速简便地监测外周血造血干/祖细胞的变化,为判断最佳采集时机提供了一种快捷简便的参考方法。
