摘要
目的观察高+G_x暴露对猴肾脏的损伤作用及恢复情况,为载人航天型号任务中可能遇到的高过载医学评价以及保障高过载人体试验安全提供依据。方法18只健康雄性成年恒河猴被随机分为对照组(n=2)、+15 G_x组(n=6)、+18 G_x组(n=3)和+21 G_x组(n=7),分别在离心机上承受相应峰值的抛物线型过载曲线作用,然后于不同时间进行尿检和解剖取材,用光学显微镜观察肾脏的病理学改变。结果①+G_x暴露后即刻,+15 G_x组、+18 G_x组和+21 G_x组猴分别出现了不同程度的尿潜血阳性、尿蛋白阳性,+21 G_x组出现尿潜血阳性的频率显著高于+15 G_x组(P<0.05),其阳性程度也高于对照组和+15 G_x组(P<0.01);②+G_x暴露后即刻,+15 G_x组、+18 G_x组和+21 G_x组猴肾脏分别出现了轻微、中度和较严重的病理损伤;③+15 G_x组、+18 G_x组猴肾功能改变和肾脏病理损伤在过载暴露后9 d基本恢复正常,+21 G_x组30 d基本恢复正常。结论高+G_x暴露可引起猴肾脏可恢复性损伤,且随着G值增加,损伤加重,恢复时间延长。
Objective To observe the pathological effect of sustained +Gx exposures on kidney in rhesus monkeys so as to provide reference for medical evaluation and health protection against high +Gx exposures possibly encountered in manned space flight and ensuring subjects' safety in high +Gx experiments. Methods Eighteen healthy male adult rhesus monkeys were randomly divided into control group(n=2) and 3 experimental groups: + 15 Gx group (n: 6), + 18 Gx group(n= 3) and +21 Gx group (n=7). They underwent +Gx exposure: +1 Gx 200 s, +15 Gx200 s, +18 Gx 165 s and +21 Gx 140 s respectively with parabolic profile on an animal centrifuge. After +Gx exposures, urine samples were collected and tested for occult blood and protein at different time points. Kidneys of 9 monkeys were dissected at different time points after exposure for gross inspection and microscopic examination. Results ①Immediately after + Gx exposures, different degrees of positive urine protein or occult blood were shown in animals of +15 Gx, +18 Gx and +21 Gx groups. The incidence of hematuria in +21 Gx group was significantly higher than that in +15 Gx group (P〈0. 05), and its positive degree was also significantly higher than that in control and +15 Gx groups (P〈0. 01). ②Immediately after +Gx exposures, slight, medium and relatively severe pathological injuries of kidney were found in monkeys from +15 Gx, +18 Gx and +21 Gx groups respectively. ③Pathological changes of kidney from monkey of +15 Gx and +18 Gx groups recovered 9 days after exposures, while in +21 Gx group recovery took place 30 days after exposures. Conclusions High sustained +Gx exposures can induce restorable functional changes and pathological injuries of kidney in rhesus monkeys. Along with the increase of +Gx load level, the severity of kidney injury increased and the time for fully recovery prolonged.
