三种玻璃化冷冻法保存小鼠早期囊胚的效果观察

摘要 目的比较三种不同的冷冻载体玻璃化冷冻保存小鼠早期囊胚,复苏后胚胎的存活率及体外继续发育能力。方法分别应用普通麦管、CPS及冷冻环玻璃化冷冻小鼠早期囊胚,以同期未冷冻早期囊胚作对照,观察冷冻复苏后胚胎存活率及继续发育能力。结果复苏后胚胎丢失率:冷冻环组显著低于普通麦管组和CPS组(P<0.05);存活率:冷冻环组和CPS组均高于普通麦管组,差异有显著性意义(P<0.05);囊胚孵出率:冷冻环组高于CPS组,二者之间有显著性差异(P<0.05),冷冻环组与对照组(同期新鲜未冷冻的早期囊胚)比较无显著性差异(P>0.05)。结论冷冻环能明提高囊胚的玻璃化冷冻效率。 Objective Three different methods of cryopreservation were compared for their ability to support post thaw survival and in vitro development of mouse early blastocysts. Methods Mice early blastocysts were vitrified by three methods straw, CPS and cryoloop vitrification. After thawing, the effects of the vitrified mice blastocysts were assessed by survival, hatched, and the lost rate. At the same time, some fresh expanded blastocysts were made as in control group. Results The lost rate of post thaw embryos vitrified in cryoloop was the lowest compared to the straw group and the CPS group ( P 〈 0.05 ), The survival rate post thaw embryos vitrified in the straw group was the lowest (P 〈 0.05). The hatched rate in the cryoloop group was higher than that in the CPS group (P 〈 0.05) , and was similar to control group (P 〉 0. 05) . Conclusion Vitrification with cryoloop can improve the efficiency of cryopreservation of early blastocysts.
