姜黄素对人舌鳞癌Tca8113细胞株增殖抑制机制的研究 被引量:1

摘要 目的探讨姜黄素(curcumin)对人舌癌Tca8113细胞株体外增殖及凋亡的调控作用。方法选用人舌鳞癌Tca8113细胞系进行体外培养,以5—40μmol/L的姜黄素分别处理Tca8113细胞24~72h后,倒置显微镜观察细胞形态,MTT法检测细胞增殖,SABC免疫组化海检测细胞内核增殖抗原ki-67、癌基因蛋白P-21、细胞凋亡相关基因蛋白Bax表达水平。结果eurcumin对Tca8113细胞增殖有抑制作用,并随药物浓度升高和时间延长而增强,倒置显微镜观察可见细胞发生凋亡形态学改变的程度和浓度与时间呈正相关,免疫组织化学检测到核增殖抗原ki-67、P-21表达均下调,Bax表达增强。结论姜黄素对Tca8113细胞的增殖具有明显抑制作用,并与浓度、时间有量效关系。通过上调凋亡基因蛋白Bax,下调ki-67、P-21蛋白的表达,诱导细胞凋亡可能是其作用机制之一。 Objective To explore the mechanism of anti - tumor effects of curcumin on human tongue cancer cell(Tca8113 ). Methods MTT colorimetry method, SABC immunohistochemistry were used. Under the effect of the curcumin, cell grew against the wall and suspended in the culture liquid, the Tca8113 cell nucleolus were found fragmentated into different size of apoptosis body under fluoroscopy. The cell proliferation were obviously suppressed after treated with different concentration curcumin for 24 ~ 72 hours. Results The apoptosis induced by curcumin of Tca8113 cell is concentration dependent and time dependent. The expression of ki -67 and p -21 in Tca8113 cell treated with curcumin down -regulated significantly, while the level of bax Curcumin can suppress Tca 8113 cell growth. The suppression up -regulated significantly. Conclusion effect is concentration and time dependent, through the pathway of downing - regulated the expression of ki - 67 and p - 21and upping - regulated bax level perhaps is one of the mechanismes inducing Tca 8113 cell apoptosis.
