热休克蛋白70与经瞳孔温热疗法实验研究 被引量:1

摘要 目的探讨经瞳孔温热疗法(TTT)对眼组织热休克蛋白70(HSP70)表达的影响。方法利用恒河猴做动物模型。于治疗后不同时间点(1天,1周,2周,1个月)摘除双眼。应用组织学及免疫组化技术,研究TTT治疗对HSP70的影响。结果TTT治疗后,于1天起,在强反应光斑边缘视网膜,脉络膜有热休克蛋白70的表达,一月时视网膜全层萎缩,激光斑边缘仍可见HSP70表达,TTT治疗可以引起视网膜不同程度组织病理学的损害。结论经瞳孔温热疗法可以引起恒河猴视网膜组织病理学损害。局部温度的升高会诱导猴眼视网膜脉络膜内源性HSP70的产生,从而提高细胞的耐热能力,达到减少组织细胞损伤的目的。 Objective To assess the effect of retinochooridal overexpression of heat shock proteins ( HSP - 70) induced by transpupillary thermotherapy on monkey model. Methods Two Henghe - Monkey were anesthetized and TTT was performed on both eye using a 810nm diode laser. At different time course after laser irradiation, a histological study was performed and immunohistochemistry was done with HSP -70 antibody on paraffin section of the eye. Results At power of 180 - 220mw, there is a faint white or white spot of laser burn. From one day to four weeks, there is strong HSP70 expression on both retinal and choreidal layer, including capillary endothelial cells. At 4 weeks, the whole layer of retina, were atrephy on the site of laser bum, Conclusion This study shows TTT can cause histology damage on retinal layer. Retinochoreidal overexpression of HSP70 can be induced after TTT on monkey model.
