摘要
目的研究固相化抗体与流动溶液中可溶性抗原结合的特点,探讨采用液相免疫捕获法自生物体内采集某些特殊样本的可行性。方法将抗体包被的ELISA反应板孔条改为样品捕获池,建立模拟生物标本(如血液)流动采集系统。以人血清IgM的ELISA检测为实验模型,观察固相抗体对流动状态下可溶性抗原捕获的特征,并与静止及适当振荡条件下抗原抗体的结合特征进行比较。结论固相化抗体对流动标本中可溶性抗原的捕获特征与静置法及震荡法基本相当,综合能力强于或显著强于后两者。
Objective To reveal the conjunct characteristics of immobilization of antibody binding to soluble antigen of mobile phase and explore the feasibility of collecting some samples from organism by liquid phase immunoprecipition. Methods To refit the ELISA reactive wells coated with antibody as the capturing pools of samples and construct the mobile collective system of the simulate biology sample (eg,blood). To detect IgM in the serum by ELISA as the experimental model, to observe all kinds of capturing characteristics of immobilization of antibody binding to soluble antigen in the mobile phase and compare its conjunct characteristics with that in the state of still and oscillation. Conclusion Compared to the capturing characteristics in the state of still and oscillation ,the capturing characteristics of immobilization of antibody binding to the soluble antigen in the mobile samples are approximately equivalent ,while the combination abilities are stronger or significantly stronger.
