摘要 [目的]观察大豆异黄酮(SI)对大鼠肾脏局部辐射后造血和抗氧化能力的影响,为探讨新型的肾脏局部辐射防护策略提供理论依据。[方法]将SD雄性大鼠48只,分为正常对照组(Non-Rad组)、辐射组(Rad组)、大豆异黄酮治疗组(SI组)和氨磷汀处理组(AFT组)。手术暴露大鼠双侧肾脏,铅板屏蔽其他组织,应用15 Gy137Csγ射线一次性照射除Non-Rad组外其余3组大鼠双侧肾脏。其中SI组从照前7天至照后14天每天1次灌胃大豆异黄酮100mg/kg,AFT组在照前30min单次腹腔注射氨磷汀200mg/kg。各组分别于照射前、照射后3天、照射后14天计数外周血WBC;于照射后7天取血,14天处死取肝脏和肾脏,观察肝肾组织的抗氧化指标变化。[结果]Rad组外周血WBC急剧下降,丙二醛(MDA)水平明显上升,超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性显著下降(P<0.05);补充SI和照射前预防性使用AFT均可显著减轻大鼠肾脏局部受照后外周血WBC下降幅度,降低肝脏MDA水平,提高红细胞数和肝脏SOD活性以及肝脏和肾脏GSH-Px活力,差异有显著性(P<0.05)。[结论]大豆异黄酮能有效缓解肾脏局部辐射引起的造血及抗氧化功能损伤,提高机体抗辐射能力。 [ Objective ] To study the effects of soybean isoflavones ( SI )on hematogenesis and antioxidation in kidneyradiated rats, to provide theoretical basis for developing new effective radioprotective drug for local radiation damage on kidney. [ Methods ] Forty-eight male SD rats were randomly assigned into 4 groups evenly, i.e. normal control group( Non-Rad group ), radiation group ( Rad group ), SI supplementation group ( SI group ) and amifostine treatment group ( AFT group ). All rats except Non-Rad group were administrated one-off 15 Gy137Cs γ-ray radiation on both kidneys through local operation with a special shield mould. SI group rats were fed with soy isoflavones 100mg/kg daily from 7 days before radiation to 14 days after, AFT group rats were given amifostine 200 mg/kg through single intraperitoneal injection 30 minutes before radiation. The changes of peripheral white blood cell were counted before radiation, on the 3rd days and the 14th days after. The antioxidative indices were observed in blood RBC on the 7th day after radiation, in liver and kidney on the 14th day after radiation. [ Results ] After irradiation to kidney in rats, there were a rapid reduction of WBC, significant increase of MDA level and depression of SOD and GSH-Px( P 〈 0.05 ). Compared with Rad group, SI group and AFT group were significantly characterized by less reduction of WBC, lower MDA level in liver, higher SOD activities in RBC and liver, and higher GSH-Px activities in liver and kidney respectively( P 〈 0.05 ). [ Conclusions ] Supplementation of soy isoflavones had significant benefits on hematopoietic system, antioxygenic property of local radiated rats, thus enhance the anti-radiation ability.
