漂白剂亚硫酸钠对小鼠生殖毒性的影响 被引量:2

摘要 [目的]研究亚硫酸钠(Na2SO3)对小鼠精子畸形及胚胎生长发育的影响。[方法]①取25只雄性昆明种小鼠随机分成阴性对照组、阳性对照组和3个剂量组,每组5只。分别按高、中、低剂量(130,65,32.5mg/kg)Na2SO3灌胃,阴性对照组用等量生理盐水灌胃,阳性对照组用20mg/kg的环磷酰胺腹腔注射。每天灌胃(注射)4次,连续5d,第35天处死动物,取两侧附睾液涂片,观察小鼠精子畸形。②将成年昆明种小鼠按雌雄2:1同笼交配。检出受精鼠,随机分成4组,在孕期第6天分别经口灌服高、中、低剂量Na2SO3(162.5,40.63,10.16mg/kg),连续9d,每天1次;阴性对照组用等量生理盐水;每3天称重一次,记录孕鼠体重变化。③在分娩前1天取出胎鼠,记录其体重、活胎数、吸收胎数、身长、尾长、胎鼠外观畸形和内脏畸形等指标。[结果]与对照组相比,各剂量组小鼠精子畸形率明增加(P<0.05),并有剂量-反应关系;孕鼠体重和胎鼠身长、体重均明显减少(P<0.05)。[结论]亚硫酸钠对小鼠精子畸形、胎鼠身长和体重具有明显影响,表明亚硫酸钠具有一定生殖毒性。 [ Objective ] To investigate the effects of sodium sulfite( Na2SO3 )on mice sperm abnormalities and embryonic development to determine whether it is a reproductive toxin. [ Methods ] ① Twenty-five mice were randomly divided into five groups ( negative control, positive control and three experimental groups ) with 5 mice per group. The positive control group was administrated intraperitoneally with cyclophosphamide at the dose of 20 mg/kg, the experimental groups were administrated with Na2S03 by gastric perfusion at low, medium and high dose respectively, the negative group was administered with saline by gastric perfusion, all administrations were once a day for 5 days successively. On the 35th day, all mice were killed and spermatozoa were obtained for observing the rate of sperm abnormalities. ② Female and male mice were copulated by the ratio of 2 : 1 to collect pregnant mice. The pregnant mice were then divided randomly into four groups ( control, low, medium and high dose ). On the 6th day of gestation, the experimental groups were administrated with Na2SO3 by gastric perfusion at low, medium and high dose respectively, the negative group was administered with saline, once a day for 9 days successively. The body weight of the pregnant mice was recorded every 3 days.③On the last day before delivery, the fetuses were obtained for observing the number of living, dead and absorbed fetuses and also skeleton abnormality, organ abnormality, weight of body, and length of body and tail. [ Results ] Sperm abnormalities in experimental groups increased significantly ( P 〈 0.05 ) compared with control group; the body weight and tail length of fetuses and the maternal body weight decreased significantly ( P 〈 0.05 ). [ Conclusion ] Sodium sulfite has reproductive toxicity on mice.
