大鼠脑挫伤后HIF-1a蛋白表达的法医学研究

Study on the expression of HIF-1a protein after cerebral contusion in rats and its forensic implication.

摘要 目的探讨大鼠脑挫伤后HIF-1a蛋白表达的规律。方法参照Feeney,s法建立大鼠脑挫伤模型,分别于伤后2h,6h,12h,24h,2d,3d,5d,7d,14d取脑,运用免疫组化SABC法观察HIF-1a蛋白的表达。结果对照组偶见阳性反应细胞。损伤后2h,即可观察到HIF-1a蛋白表达,6h后表达明显增强,24h达高峰,随后阳性反应细胞逐渐减少,7d后仍可见少量表达,14d后恢复。表达呈弥漫性,以大脑皮层、海马、脑干最为明显,不同部位的变化趋势一致。各实验组与对照组比较有统计学意义(P<0.05)。结论HIF-1a蛋白的表达规律可用于脑挫伤的鉴定及损伤经过时间的推断。 Objective To explore the expression of HIF-1α protein in different time after cerebral contusion (CC). Methods Based on Feeney's model of CC and took the samples at 2h,6h,12h,24h,2d,3d,5d,7d,14d after brain injury.The expression of HIF-1α protein was observed by immunohistochemical SABC and image analysis techniques. Results The expression of HIF-1α protein could be detected at 2h after injury, increased at 6h and peaked at 24h.Positive cells decreased with the prolongation of injury time.Little expression was found at 7d. The control group and the 14d group showed negative staining.The expression of HIF-1α protein was found in wide range but significant in cortex, hippocampus and brain stem. The changing tendency of different areas was same. Conclusion Expression of HIF-1α protein is correlated with post-traumatic intervals after CC in rats, suggesting that the expression of HIF-lot may be served as the markers for timing of CC in forensic practice.
