摘要
目的检测肿瘤坏死因子相关凋亡诱导配体(tumor necrosis factor-related apoptosis-inducing ligand,TRAIL)及抑制凋亡基因家族成员survivin在胃癌组织中的表达,并探讨其意义。方法利用半定量逆转录聚合酶链反应(RT-PCR)法检测82例胃腺癌及对应的正常胃组织中TRAIL和survivin的表达。结果TRAIL在正常胃组织中的表达阳性率及相对表达量均明显高于癌组织(P<0.05);而survivin的表达则与TRAIL表达相反,正常胃组织中不表达,在胃癌组织中的表达阳性率为62.2%。TRAIL和survivin表达量与肿瘤的分化程度有关,分化程度越低,TRAIL mRNA表达量越低,而survivin mRNA表达量越高,高分化胃腺癌与低分化胃腺癌间差异有统计学意义(P<0.05)。结论TRAIL和survivin在胃癌凋亡过程中起着重要的作用,TRAIL可诱导凋亡,survivin则抑制凋亡发生。
Objective To investigate the expression of tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) and survivin in gastric carcinoma. Methods Expression of TRAIL and survivin were assayed by semiquantitive reverse transcriptase polymerase chain reaction (RT-PCR) in 82 cases of gastric carcinoma and normal gastric tissues. Results The positive rate and expression quantity of TRAIL in normal tissues were higher than those in cancer tissues( P 〈0.05). However,the survivin was only expressed in cancer tissues,the positive rate was 62.2%. The expression quantity of TRAIL and survivin was correlated with histological grade, the expression of TRAIL was higher in well-differentiated gastric adenocarcinoma than that in poorly differentiated gastric adenocarcinoma while the expression of survivin was lower( P 〈 0.05 ). Conclusion TRAIL can induce apoptosis and survivin can depress the happening of apoptosis,which may be one of the mechanisms by which gastric carcinoma develops.
