摘要
目的:研究灌服赤芍水提物后的犬药物血清作用于肝星状细胞株HSC-T6基因蛋白表达的变化。方法:采用乙醛造模后的肝星状细胞株HSC-T6细胞作为酒精性肝纤维化的体外研究模型;以赤芍水提物给彼格犬一次性灌胃,取给药后2小时的血清作为实验药物血清。以免疫细胞化学法检测药物血清对HSC-T6作用72小时后bax、bcl-2、caspase-33种基因蛋白表达情况,并与乙醛造模后的HSC-T6细胞相比较。结果:乙醛造模后的HSC-T6细胞中bcl-2的表达较正常培养组细胞增加,bax、caspase-3表达降低;药物血清作用后的HSC-T6细胞中bcl-2的表达较造模组的HSC-T6细胞显著降低,bax、caspase-3的表达较模型组细胞显著增加。结论:赤芍水提物入血后可能是通过bax、bcl-2、caspase-3等基因异常表达来实现对HSC-T6的抑制增殖及促进凋亡作用。
Objective: To study the effects of dog serum after being fed with Chishao distilled substances on hepatic stellate cell (HSC-T6) in vitro, trying to analyze the protein levels of bax, bcl-2, caspase-3 in HSC-T6. Methods: The experiments adopted HSC-T6 builded model by acetaldehyde as the model for study in vitro. Feeding the dog with Chishao distilled substances, took the serum at 2-hour point after feeding as the medicated serum for experiment. By immunohistochemistry method, the protein levels of bax, bcl-2, caspase-3 in HSC-T6 were analyzed. Results: The results showed that compared with common HSC-T6, builded model HSC-T6 increases of bcl-2 proteins were observed, while decrease of bax, caspase-3 protein levels was obtained. Compared with model, HSC-T6 functed medicated serums under the same conditions, decrease of bcl-2 proteins were observed, while increases of bax, caspase-3 protein levels was obtained. Inhibition rate was the highest at the 2-hour and 3- hour medicated serum. Conclusion: These results lead to a conclusion that exposure Chishao medicated serums can change the expression of apoptosis-related genes, and it suggests that Chishao medicated serums can induce apoptosis in HSC-T6 and may be related with some apoptosis-related gene such as bax, bcl-2, and caspase-3.
