摘要
目的:既往研究表明,染色体8p21区与精神分裂症连锁,位于8p21.3-p22区的成纤维细胞生长因子20(fibroblast growth factor,FGF20)基因可能与精神分裂症关联,本研究旨在探讨FGF20基因与汉族人群精神分裂症的关联。方法:本研究采用聚合酶链式反应—限制性片断长度多态性方法,分析301例精神分裂症患者和319例正常对照者中FGF20基因5’端单核苷酸多态性rs1721100与精神分裂症的关联。结果:与正常对照组相比,精神分裂症患者rs1721100多态性的基因型(x2=21.977,df=2,p<0.001)和等位基因(C>G,x2=5.249,df=1,p<0.001,OR=1.67,95%CI:1.32-2.11)的频率差异有显著性。结论:本研究结果提示FGF20基因遗传多态性可能与精神分裂症存在关联。
Objective: Previous studies have reported genetic linkage evidence for a candidate gene of schizophrenia on chromosome 8p21, Recently, it has been striking research hotspot to explore the role of fibroblast growth factor 20 (FGF20, 8p21.3- p22) in neurodevelopmental process, In the present study, we proposed to explore the association of the single nudeotide polymorphism (SNP) rs1721100 at the 5' region at FGF20 locus with schizophrenia in Chinese Hart population, Methods: The genotypes of 301 schizophrenic patients and 319 healthy controls were examined by using the polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). Deviation of the genotype counts from the Hardy-Weinberg equilibrium was tested using a chi-square goodness-of-fit test. With the chi- square tests, the frequencies of genotypes and alleles of this polymorphism were compared between the cases and control groups. Results: Genotype frequencies of the SNP rs1721100 in schizophrenia and healthy control groups did not show significant deviations from Hardy Weinberg equilibrium. Compared with the healthy control group, the schizophrenic patients showed significant difference in frequencies of both genotypes (x^2=21.977, df=2, p〈0.001 ) and alleles (C〉G, x^2=5.249, df=1, p〈0.001, OR=1.67, 95%CI: 1.32-2.11 ) of SNP rs1721100. Conclusion: Our results suggest that the SNP rs1721100 at the FGF20 gene is likely to play a major role in the susceptibility to schizophrenia in Chinese Han population.
