摘要 对海滨锦葵组织培养再生体系进行了研究,结果表明:1.海滨锦葵茎段外植体较好的愈伤组织诱导培养基组合为MS+KT1.5mg/L+IAA2.0mg/L或MS+6-BA4.0mg/L+IBA0.2mg/L+Inositol50mg/L.2.海滨锦葵愈伤诱导不定芽的较好培养基组合为MS+NAA2.0mg/L+2iP5.0mg/L和MS+NAA0.5mg/L+2iP5.0mg/L.3.海滨锦葵腋芽分化不定芽的较好培养基组合为改良MS(含2倍有机营养)+KT0.4mg/L+IAA1.0mg/L.MS基本培养基中适当提高有机营养的含量对腋芽的增殖有促进作用,实验中以有机营养的含量为基本培养基2倍时效果较好.4.不定芽在1/2MS的培养基中附加IBA1.0mg/L生根最好. The study on the regeneration of in vitro culture stems and adventitious shoots from axillary buds or top buds of Kostelezkya virginica was conducted. The stems were wounded by stem cuts and cultured on media based on Murashige and Skoog (MS) basal medium, containing different plant growth regulators with different concentration combinations. The stem callus were induced with higher frequency from the surface of stems as explants when cultured on MS basal medium supplemented with 1. 5 mg/L kinetin (KT) and 2. 0 mg/L indole-3 acetic acid (IAA), or on MS basal medium supplemented with 4. 0 mg/L 6-benzyladenine (6-BA), 0. 2 mg/L indole- 3-butyric acid (IBA) and 50 mg/L inositol. The optional medium for adventitious shoot induction from callus were MS basal medium supplemented with 2. 0 mg/L Naphthalene (NAA) and 5.0 mg/L 2-Isopentyladenine (2iP), or supplemented with 0. 5 mg/L NAA and 5.0 mg/L 2iP. The optional medium of proliferation of axillary buds or tip shoots was the improved MS medium (2X organic nutrition instead of 1 × organic nutrition) supplemented with 0. 4 mg/L KT and 1. 0 mg/L IAA. The higher concentration of organic supplements seemed to improve the micropropagation of regeneration shoots. The improved MS basal medium promoted the proliferation and growth of regenerated shoots. The healthy adventitious buds and regenerated shoots could be rooted well on 1/2 MS medium supplemented with 1.0 mg/L IBA.
