摘要
目的制备紫杉醇磁性脂质体。方法采用薄膜超声法制备紫杉醇磁性脂质体;用透射电镜考察脂质体形态;高效液相色谱法测定紫杉醇磁性脂质体紫杉醇含量及组织分布。结果制得紫杉醇磁性脂质体外观、形态良好,平均粒径600-800 nm,包封率在90%以上。紫杉醇磁性脂质体体内、外具有良好得磁场响应性。应用紫杉醇磁性脂质体加磁场方式给药可以使紫杉醇有效的聚集到靶部位。结论紫杉醇磁性脂质体在磁场的作用下可以有效提高组织内化疗药物浓度。
Objectives To prepare paclitaxel magnetic liposomes. Methods Paclitaxel magnetic liposomes were prepared by means of film ultrasound an its morphology was examined with transmission electron microscope. Paclitaxel magnetic liposomes and tissue distribution were quantified with high-performance liquid chromatography (HPLC). Results We obtained the paclitaxel magnetic liposomes with good appearance. The size of particles was about 600-800 nm, the encapsulation rate being above 90 %. Paclitaxel magnetic liposomes have a good response to the magnetite both outside and inside the body. Administration of paclitaxel magnetic liposomes under magnetic field could be used to deliver paclitaxel effectively to the targeted site. Conclusion Paclitaxel magnetic liposomes could be used to deliver paclitaxel effectively to the tumor with a permanent magnet.
