复方壳多糖组织工程皮肤分泌b-FGF的初步研究 被引量:1

摘要 目的研究组织工程皮肤培养液中碱性成纤维细胞生长因子(b-FGF)的分泌情况。方法收集复方壳多糖组织工程皮肤制备过程中复方壳多糖皮肤替代物(SE)、复方壳多糖真皮替代物(DE)、单层角质形成细胞(KC)以及复方壳多糖DE与KC在transwell内共培养的培养液样本,双抗体夹心ELISA法测定其中b-FGF分泌量。结果b-FGF分泌量依次为单层KC<复方壳多糖DE/KC共培养<复方壳多糖SE<复方壳多糖DE。结论角质形成细胞能调控成纤维细胞分泌b-FGF能力,有利于伤口愈合。 Objective To investigate the secretion of basic fibroblast growth factor(b-FGF) in cultured tissue-engineered skin. Methods b-FGF from the composite chitosan skin equivalent(SE), composite chitosan dermal equivalent(DE), monolayer keratinocyte(KC) ,and DE/KC co-cultured in transwell were detected by ELISA. Results b-FGF from SE was less than that from DE, but was more than that from DE/KC co-cultured,and monolayer keratinocyte secreted the least b-FGF. Conclusion Keratinocyte can control fibroblast's secretion of b-FGF,which benefits the wound healing process.
