摘要
目的观察牛磺酸对过度训练大鼠心肌细胞凋亡及fas和caspase-3基因表达的影响,探讨牛磺酸对运动心脏保护作用的可能机制。方法采用力竭性游泳训练建立大鼠过度训练模型,30只SD大鼠随机等分为正常对照组、过度训练组和过度训练+牛磺酸组。实验后采用流式细胞仪检测各组大鼠心肌细胞凋亡,半定量RT-PCR检测caspase-3和fas mRNA表达,二步法免疫组化检测Fas和Caspase-3蛋白表达。结果过度训练组大鼠心肌细胞凋亡率、fas和caspase-3基因表达水平均显著高于正常对照组(P<0.01)。而过度训练+牛磺酸组心肌细胞凋亡率、fas和cas-pase-3基因表达与正常对照组无显著差异(P>0.05)。结论牛磺酸对过度训练大鼠心肌细胞凋亡以及fas和caspase-3基因表达增高有较好的颉颃作用。
Objective sion of fas and caspase - 3 To observe the effects of taurine on the apoptosis of cardiomyocyte and the expresgenes in overtrained rats, and to explore the possible mechanism of taurine in protection of cardiomyocyte damage from excessive exercise. Methods Overtrained rat models were established through exhaustive swimming. Rats were randomly divided into the control group(Group A), excessive exercise group(Group B) and excessive exercise + taurine group(Group C). The rate of apoptosis was determined by flow cytometry. The expression of fas mRNA and caspase - 3 mRNA were detected by semi - quantitive RT - PCR. The expression of fas and caspase - 3 proteins were examined by two - step immunohistochemical staining. Results The rate of apoptosis, the expression of fas and caspase - 3 genes in the myocardial cell in group B were significantly higher than that in group A(P 〈 0.01). There were no significantly difference in the rate of apoptosis, the expression of fas and caspase - 3 genes in myocardial cell in group C as compared with that in group A( P 〉 0.05). Conclusion Taurine exerts a better antagonist action on the apoptosis of myocardial cells and the expressions of fas and caspase- 3 genes in overtrained rats.
