摘要
目的:观察FY-10对无血清培养HaCat细胞分化的影响。方法:选择不同浓度的FY-10(10^-5~10^-9mol/L)分别添加于无血清培养的HaCat细胞培养液中,并在添加后24h回收总RNA,用逆转录PCR(RT—PCR)方法检测Hacat细胞分化的特异性标记TGase1、KRT6的mRNA表达情况。结果:FY—10添加后24h能显著的上调TGasel的mRNA的表达,其上调的程度与药物浓度成良好的线性关系;而FY-10对KRT6的mRNA表达呈显著的抑制作用,呈一定的剂量依赖性。结论:一定浓度下FY-10能上调无血清培养HaCat细胞对分化特异性标记蛋白TGase1mRNA的表达,而对KRT6mRNA有下调其表达的作用,促进HaCat细胞的分化,而抑制其增殖。
Objective: To determine the effect of FY- 10 on the differentiation of HaCat cells cultured in serum free medium. Methods: Various doses of FT- 10 (10^-5 - 10^-9 mol/L) were added into confluent HaCat cells cultured in serum free medium. Twenty - four hours after adding FY - 10, total RNA was abstracted to detect the expression of specific keratinocyte differentiation markers. The expressions of transglutaminase 1 and keratin 6 were measured by RT- PCR method. Results: Transglutaminase 1 was up- regulated apparently in a dose - dependent manner. However, keratin 6 was down - regulated in a dose - dependent manner. Conclusion: FY - 10 can induce the differentiation via up- regulation of transglutaminase and inhibit the proliferation of HaCat cells.
