摘要
目的通过乙醇灌胃的方法造成生长期小鼠骨丢失的模型,探讨少量乙醇对生长期小鼠骨生物力学性能的影响。方法清洁级6周龄昆明种小鼠30只,体重25~35g,雌雄各半,随机分成3组,(1)基础对照组,实验开始时处死取材;(2)正常对照组,给予蒸馏水10 m1·kg^(-1·d-1)灌胃:(3)乙醇组,给予50%(V/V)乙醇(根据预实验结果选用),按4 g·kg^(-1·d-1)剂量灌胃,换算成60 kg体重成人相当于摄入乙醇约30 g/d,连续灌胃60d。实验结束时,眼球取血处死,取材:第4腰椎做凹入实验、右侧股骨做侧摔实验。结果腰椎最大抵抗应力降低(-63.6%,P<0.01),股骨颈结构强度下降(-29.4%,P<0.01)。结论持续少量乙醇摄入可造成生长期小鼠的腰椎和股骨颈的力学性能下降。
Objective We produced a model by given alcohol orally in order to study the effects of little ethanol on bone biomechanics property of growing mice. Methods Thirty six-week-old KM mice, clean grade, half male and half female, initially weighing 25-35 g, were randomly divided into 3 groups. These groups were basel control which were killed at the beginning of the experiment; intact control that were given distilled water with 10 ml·kg^-1·d^-1, 50%(V/V) ethanol group (Model) according to the dosage of 4 g·kg^-1·d^-1 for 60 days. All mice were killed by taken out their eyeball to collect the forth lumbar vertebra and right femur for testing biomechanical properties including Indentation Test of vertebra and fall-loading test of proximal femoral neck. Result Maximal resistibility of lumbar vertebra decreased 63.6% significantly (P〈0.01) and structural mechanical stranth of right proximal femoral neck declined (-29.4%, P〈0.01) in Model group, Conclusion Little ethanol had badly affected biomechanical properties including vertebra and proximal femoral neck in growing mice.
