摘要 应用SRAP对金针菇种质资源进行评价.选用多态性好的7对SRAP引物对30个来源不同、各具代表性的供试菌株进行PCR,共获得92条重复性良好的DNA条带,其中多态性条带79条,占85.9%;用SPSS软件计算相似系数和进行平均法聚类分析,白色菌株之间遗传差距较小,黄色菌株之间遗传差距大,30个供试菌株可分为14类.本研究还表明,SRAP具有高效、稳定、重复性好的特点,可应用于遗传连锁群构建、品种鉴别及分子标记辅助育种等研究. Sequence-related amplified polymorphism (SRAP) was used to evaluate the germplasm resources of Flammulina velutipes. Seven pairs of primers with good polymorphism were selected to amplify 30 representative F. velutipes strains from different sources. Ninety two repeatable DNA bands from tested F. velutipes were obtained, of which 79 were polymorphic, accounting for 85.9%. The coefficient of similarity was calculated and duster analysis was conducted with SPSS software, and it was proved that the genetic difference between yellow strains was more significant than that between white strains. The 30 tested F. velutipes strains were classified into 14 categories. It was also showed that SRAP analysis was efficient, stable and repeatable in this study, and could be used in the construction of the genetic linkage map, identification of strains and molecular marker assisted breeding.
