摘要 目的观察重组C反应蛋白(CRP)对人脐静脉内皮细胞(HUVEC)基质金属蛋白酶2(MMP-2)表达的影响。方法采用逆转录聚合酶链反应及蛋白质免疫印迹分析检测MMP-2mRNA转录和蛋白水平表达,观察不同浓度CRP及不同孵育时间对HUVEC MMP-2表达的影响。结果CRP呈浓度和时间依赖性增加培养HUVEC的MMP-2mRNA转录和蛋白水平的表达。结论CRP可上调HUVEC MMP-2表达。 Objective To investigate the effects of C-reactive protein(CRP) on expression of matrix metalloproteinase-2 ( MMP-2) in human umbilical vein endothelial cells ( HUVEC). Methods HUVEC were cocultured with different concentration of CRP for different time. MMP-2 protein and mRNA expression in HUVEC were measured by Western blotting and reverse transcription polymerase chain reaction(RT-PCR). Results Incubation of HUVEC with CRP results in an increasing expression of protein and mRNA of MMP-2 in a concentration and time-depedent manner. Conclusion CRP enhances the expression of MMP-2 in HUVEC.
