细胞色素P450 2E1基因多态性与结直肠癌易感性的关系

Polymorphisms of CYP2E1 Rsa Ⅰ and susceptibility of colorectal cancer

目的:研究细胞色素P4502E1(CYP2E1)基因多态性与结直肠癌易感性的关系。方法:对315例结直肠癌患者和439名对照人群进行病例对照研究,PCR-RFLP检测其基因型。结果:1)c1/c1、c1/c2和c2/c2基因型分布频度在结直肠癌组和对照组间差异有统计学意义,χ2=6.58,P=0.037。与c1/c1基因型相比,c2/c2基因型发生结肠癌的危险性显著升高;2)有饮酒习惯者发生结直肠癌的危险性显著升高;3)CYP2E1RsaⅠ多态与饮酒习惯有显著的协同作用。结论:CYP2E1RsaⅠ基因多态和饮酒习惯影响结直肠癌的易感性,二者在结直肠癌发生中有显著的协同作用。

OBJECTIVE： To study the relationship between genetic polymorphisms of cytochromes P450 Rsa Ⅰ and the susceptibility of colorectal cancer. METHODS： A case-control study was conducted with 315 cases of colorectal cancer and 439 population-based controls. Genotypes were detected by PCR-RFLP. RESULTS： 1 ） The frequencies of c1/c1 ,c1/c2 and c2/c2 genotypes distribution in coloreetal cancer group were significantly different from those of control group,Х^2=6.58, P= 0. 037. Individuals who had c2/c2 genotypes were at an increased risk of developing eolorectal cancer compared with those with c1/c1 genotypes. 2）Individuals who had alcohol drinking habit were at an increased risk of developing colorectal cancer. 3）There is a significantly cooperated effect between CYP2E1 Rsa Ⅰ polymorphism and drinking habit in developing colorectal cancer. CONCLUSION： Polymorphisms of the CYP2E1 Rsa Ⅰ and alcohol drinking influence the susceptibility to colorectal cancer,and they have the significantly synergic effect on the developing colorectal cancer,
