摘要
目的检测前炎性因子、趋化因子及其受体在小鼠角膜碱烧伤后表达的动态变化。方法建立小鼠碱烧伤模型,用RT-PCR法检测角膜碱烧伤后不同时间点前炎性因子、趋化因子及其受体mRNA的表达。结果RT-PCR检测显示,角膜碱烧伤后角膜组织中前炎性因子IL-1α、IL-1β及IL-6表达增强;调控单核细胞迁移的CCR1-CCL2、CCL3、CCL4、CCR2-CCL2以及调控中性粒细胞迁移的CXCR2-CXCL2表达增强。结论角膜碱烧伤后前炎性因子以及参与调控白细胞迁移的趋化因子及其受体明显上调,提示其参与角膜碱烧伤后白细胞向局部的迁移。
Objective Corneal neovascularization is a common manifestation of many corneal lesions,including infection and trauma. However, the pathogenesis of corneal neovascularization is unclear. We worked to define the pathologic role of proinflammatory cytokine,chemokine and its receptor in CNV and detected the gene expression in mouse cornea using alkali injury induced corneal neovascularization model. Methods The murine corneas were wounded by cautery with the filter paper containing 1 mol/L NaOH for 45 seconds in the right eyes of 45 C57BL/6 mice, and the fellow eyes were as controls. Five eyeballs were enucleated at 0,2,4,7,14,28 days respectively for histological examination. RT-PCR was used to detect the gene expression of proinflammatory cytokine, chemokine and chemokine receptor at 0,2,4,7 days after cauterization. Results Lots of leucocytes infiltration was found in cornea at 2,4 days after burned,and obvious new blood vessels were induced by alkali-burn 2 weeks. RT-PCR showed that the gene of proinflammatory cytokine ( IL-1, IL-6) and chemokine receptor and its ligand ( CCR1- CCL2, CCL3, CCL5,CCR2-CCL2 and CXCR2-CXCL2) in the injured right corneas were enhanced at day 2,4 and 7 after alkali injury compared with the control corneas (IL-6:F =5.58,P 〈0.01;CCL3:F = 8.0,P 〈0.01;CCL5:F =7.54,P 〈0.01; CXCR2 : F = 8.89,P 〈 0.01 ; CXCL2 :F = 19.94, P 〈 0. 01 ). Conclusion After alkali burn of cornea, the gene expression of proinflammatory cytokine, chemokine and its receptor increase. These factors may play an important role in mediating the infiltration of the leukocytes to the alkali injured cornea.
