摘要 目的研究前列腺素类似物的新成员Bimatoprost的降眼压作用,并评价其安全性。方法对正常无色素兔的一只眼滴入Bimatoprost,另一只眼滴入生理盐水,连续测量滴药前及滴药后的双眼眼压,绘制用药后眼压随时间变化曲线;并观察结膜充血情况及测量瞳孔直径。结果单眼点用Bimatoprost0.5h后产生显著的降眼压作用,平稳持续24h,用药3周后降眼压幅度达25%~32%。用药后结膜充血程度较轻,药物对兔眼瞳孔直径无明显影响。结论0.03%Bimatoprost可降低无色素兔眼压并有较好的安全性。 Objective Bimatoprost is a new prostaglandins,and its strongly lowing intraoeular pressure(IOP) effect on openangle glaucoma has been proved in clinical researches. The goal of this study was to evaluate the effect, safety of bimatoprost in lowing IOP in albino rabbits. Methods Twelve albino rabbits were utilized in this study. 25 pA of bimatoprost drops was topically administrated in one eye of each rabbit and sterile normal saline was used in the same way in the fellow eye. IOP of both eyes was measured before drug administration and 0. 5,1,2,5,12,14,20 and 24 hours after use of bimatoprost drops to draw the IOP curve in 4 rabbits,and the dynamic changes of IOP, status of eonjunetiva and size of pupil were recorded and assessed in 8 experimental rabbits during continuous administration of bimatoprost for 21 days. Results IOP decreased by 25% - 32% half an hour after the application of bimatoprost in albino rabbit and the effect lasted over 24 hours. During administration of drug,IOP of experimental rabbits kept a tendency of decrease for 3 weeks and the IOP was (7. 35 ± 0.46) mmHg. No significant effect of bimatoprost was found on the pupil size within 24 hours after the application of bimatoprost. Slight conjunetival hyperemia was seen in the early stage in most eyes half an hour after application of bimatoprost but alleviated after 5 hours. One week later,eonjunetival hyperemia became steady and slight. Conclusion 0.03% bimatoprost can lower IOta and is safe in normal albino rabbit eyes.
