摘要
目的观察白细胞介素-1(IL-1)对房水动力学的影响,进一步验证其降低眼压的有效性及探讨其降低眼压的机制。方法SD大鼠15只,随机分成3组,A、B组随机一只眼前房注射重组鼠IL-1β30ng,另一只眼前房注射等体积的0.1%PBS作为对照,C组作为空白对照组。用药80h后,A组用药眼和对侧眼分别前房注射10μl125I标记鼠清蛋白(125I-RSA)和131I标记的鼠清蛋白(131I-RSA),B组相反,用药眼和对侧眼分别前房注射10μl131I-RSA和125I-RSA,C组随机一只眼前房注射10μl125I-RSA,对侧眼注射10μl131I-RSA,并于注药后每10min取静脉血一次,共7次,进行血循环中125I-RSA或131I-RSA放射性强度的测定,计算血样品中125I-RSA/131I-RSA的蛋白量比值。结果三组间比值差异有统计学意义(P=0.000),A组较C组增加27.20%,B组较C组降低25.44%。结论IL-1具有增加小梁网通道房水流出的作用,为其降低眼压提供客观依据。
Objective Matrix metalloproteinases (MMPs) are proved to increase aqueous outflow facility,and interleukin-1 ( IL-1 ) is demonstrated to be a strong activator of MMPs. The purpose of the study was to ascertain the ocular hypotensive efficacy and to determine the underlying intraocular pressure(IOP) reduction mechanisms of IL-1 by studying its effects on aqueous humor dynamics. Methods Fifteen Sprague-Dawley rats were randomized into three groups. In group A and B,30 ng of IL-1β was injected intracamerally in one eye of rats ( experimental eyes) , and equivalent volume 0.1% PBS was injected intracamerally in the fellow eyes. The rats in group C were used as blank controls. Eighty hours after treatment,experlmental eyes of rats in group A received injection of 10 μl ^125I-labeled albumin intracamerally and the contralateral eyes were treated with equivalent volume of ^131I-labeled albumin. The experimental eyes in group B received 10 μl ^131 I-labeled albumin intracamerally and the contralateral eyes were treated with equivalent volume of ^125I-labeled albumin. One eye in group C was randomly treated with 10 μl ^125 I-labeled albumin and the contralateral eye was treated with equivalent volume ^131I-labeled albumin. Blood examples were harvested at 10- minute interval after albumin injections for seven times. Radioactivity of blood samples was measured by isotope counter, and the weight ratio of ^125 I-labeled albumin /^131 I-labeled albumin were calculated. ResuIts The weight ratio of ^125I-labeled albumin /^131I-labeled albuminwas 1.45 ±0. 09 in group A,0. 85 ±0. 07 in group B and 1.14±0. 10 in group C,showing a statistically significant difference among these three groups( F = 58.89 ,P = 0. 000). The weight ratio in group A increased by 27.20% in comparison with group C,and the ratio in group B decreased by 25.44% compared with group C. Conclusion IL-1 increases aqueous humor outflow through trabecular meshwork,which may be its major underlying mechanism of IOP reduction.
