原花青素交联的支架材料的生物相容性研究 被引量:1

摘要 目的探讨原花青素作为交联剂的壳聚糖、胶原蛋白、硫酸软骨素、透明质酸钠合成的支架材料植入兔角膜的生物相容性。方法将壳聚糖、胶原蛋白、硫酸软骨素、透明质酸钠等溶液按照一定的比例混合,加入原花青素和戊二醛溶液作为交联剂,制成2种支架材料分别植入2组新西兰大白兔角膜的囊袋内。在不同的时间点进行裂隙灯显微镜观察照相,并进行组织病理学检查,比较不同的支架材料对角膜的影响。结果原花青素交联组术后1周有结膜睫状充血,角膜水肿;2周后充血水肿逐渐消失。兔眼角膜情况稳定,术后4个月基本透明;材料与角膜的生物相容性好,5个月后材料大部分吸收溶解,角膜透明。组织病理学检查可见兔角膜标本中材料内有角膜基质细胞长入。原花青素交联的支架材料比戊二醛交联的支架材料刺激性小。结论原花青素交联的壳聚糖、胶原蛋白、硫酸软骨素、透明质酸钠组成的支架材料与角膜组织具有良好的生物相容性,原花青素可能比戊二醛更适合作为支架材料的交联剂。 Objective To evaluate the biocompatibility of scaffold synthesized by chitosan, collagen protein, chondroitic acid and sodium hyalurouate with proeyanidine as crosslinking reagent implanted into rabbits eye. Methods Chitosan; collagen, chondroitic acid and sodium hyalurouate were mixed with certain ratio ,then proeyanidine and glutaraldehyde were added as crosslinking reagent respectively to form two different scaffolds, which were implanted into rabbit corneal stroma respectively. Slit-lamp biomicroscopy and histopathological observation were evaluated at different time. The effect of different scaffolds on cornea was compared. Results In the proeyartidine group, the ciliary congestion and corneal edema appeared in all eyes at the first week postoperatively,and then disappeared gradually after 2 weeks;The cornea in rabbit eyes were stable and transparent basicly at 4 months; The biocompatibility of scaffolds was good, the scaffolds degenerated gradually after 5 months. Histopathologieal observation showed the corneal stromal cells grew into the scaffolds. Scaffolds with proeyanidine as crosslinking reagent had weaker stimulation than that with glutaraldehyde as crosslinking reagent. Conclusion Scaffold synthesized by chitosan,collagen protein,chondroitic acid and sodium hyalurouate with procyanidine as crosslinking reagent has good biocompatibility. Procyanidine may be more suitable as crosslinking reagent for tissue engineering scaffold than glutaraldehyde.
